The storage of cells at temperatures below -130 ºC is the temperature at which biological activity ceases. Storing at these temperatures is necessary to preserve the material unaltered. Mechanical devices are available to maintain temperatures below this temperature but they can be noisy, generate heat and generally require liquid nitrogen backup to avert disaster should the electricity supply be disrupted or the freezer fail. This makes simple liquid nitrogen cryogenic storage at -196°C a very popular choice.
To minimize the risk of contamination associated with liquid nitrogen and to ensure there can be no cross contamination with the stored samples, it is generally recommended that samples be stored in the vapour phase above the nitrogen (-146ºC).
