Just as in humans, there are two basic techniques of forensic DNA analysis used for individual identification of animal samples. The biological quality of the sample determines which technique is most appropriate. The preferred technique, STR typing, requires nuclear DNA. This means that the sample must contain a sufficient quantity of intact cells. In animals, such samples are typically bloodstains, saliva residues, and hairs with intact hair bulbs or roots. STR analysis is preferred because it is highly discriminating; a complete STR profile of an individual is virtually unique. Even an incomplete profile, common when working with hairs with marginal root material, is powerfully informative. The second technique, mitochondrial typing, is less discriminating but can be successfully performed on samples in which STR testing fails, such as hairs without roots.
